Beschreibung

General description

Interferon-g (IFN-gamma) ist ein lymphoider Faktor, der eine potente antivirale Wirkung aufweist. Es stimuliert auch Makrophagen und NK-Zellen. Menschliches IFN-gamma ist ein 16,7-kDa-Protein, das 143 Aminosäurereste enthält.

Produktherkunft: Menschlich, in E. coli exprimiert

Application

In den meisten In-vitro-Anwendungen übt IFN-gamma seine biologische Aktivität bei einer Konzentration im Bereich von 0,1 bis 10,0 ng/ml aus. Responderzellen sind unter anderem: Lymphozyten.

Physical form

Sterilfiltriert, aus 10 mM Natriumphosphat, pH-Wert 7,4, lyophilisiert.

Preparation Note

Das lyophilisierte IFN-g, wenngleich stabil bei Raumtemperatur, sollte getrocknet bei -20 °C aufbewahrt werden. Nach kurzem Schleudern mit 1x PBS, pH-Wert 8,0, auf eine Konzentration von 0,5–1,0 mg/ml rekonstituieren. Hinweis: Weitere Verdünnungen sollten in RPMI +10 % FCS oder 5 % HSA erfolgen und bei 4 °C bis zu einer Woche oder bei -20 °C langfristig gelagert werden. Rekonstituiertes Material kann in unverdünnten Aliquoten bei -20 °C bis zu sechs Monate gelagert werden.

Analysis Note

Spezifische Aktivität: Menschliches IFN-gamma ist im Vergleich zu Standards vollständig biologisch aktiv. Die durch einen Virusresistenz-Assay bestimmte ED(50) beträgt ≤ 0,05 ng/ml.
