Diabetic nephropathy is a leading cause of end-stage renal disease, characterized by endothelial dysfunction and a compromised glomerular permeability barrier. Dysregulation of the angiopoietin 1 (ANGPT1)/angiopoietin 2 (ANGPT2) signaling axis is implicated in disease progression. We recently described the discovery

Recent evidence suggests that chemokine CXCL12, the cognate agonist of chemokine receptors CXCR4 and ACKR3, reduces thrombin-mediated impairment of endothelial barrier function. A detailed characterization of the effects of CXCL12 on thrombin-mediated human lung endothelial hyperpermeability is lacking and structure-function

Questions

1–3 of 3 Questions
  1. Is it possible to separate inserts and use them in different experiments? Or should the whole plate be used at once?

    1 answer
    1. The In Vitro Vascular Permeability Assay is performed in a 96-well receiver plate with 96 cell culture inserts connected in a single tray unit. Because the inserts are interconnected, the risk of cross-contamination is significantly increased if only a subset of wells is used at a time. In addition, the collagen coating of unused wells may be compromised during incubation. For these reasons, we strongly recommend utilizing all inserts within a single experiment to ensure assay integrity and reproducibility.

  2. Could you provide information on the pore size of EMD Millipore ECM644?

    1 answer
    1. The pore size of ECM644 is 1.0 µm. It is a 6.5 mm diameter filter with a filtration area of 0.33 cm², which serves as the growth surface area.

  3. Could you provide the pore size information for ECM642?

    1 answer
    1. The ECM 642 filter has a pore size of 1.0 µm, a filter diameter of 0.4 mm, and a filtration area of 0.12 cm² (growth surface area).
