General description

B834 ist der Elternstamm für BL21 (1). Diese protease-defizienten Wirte sind Methionin-Auxotrophe und ermöglichen eine hochspezifische Markierung von Zielproteinen mit 35S-Methionin und Selenomethionin für die Kristallographie (2).

DE3 weist darauf hin, dass der Wirt ein Lysogen von λDE3 ist und daher eine chromosomale Kopie des T7-RNA-Polymerase-Gens enthält, das durch den lacUV5-Promoter kontrolliert wird. Solche Stämme eignen sich zur Herstellung von Protein aus Zielgenen, die in pET-Vektoren kloniert werden, wobei IPTG als Induktor verwendet wird.
