Beschreibung

General description

Eine Chinazolinverbindung, die als spezifischer und starker Inhibitor der Autophagie wirkt und den Abbau des Vps34 PI 3-Kinase-Komplexes fördert, indem sie die Aktivität der deubiquitinierenden Enzyme USP 10 (IC50 = 580 nM) und USP 13 (IC50 = 690 nM) blockiert. Es hat jedoch keinen Einfluss auf die Lipid-Kinase-Aktivität Vps34. Es hat sich auch gezeigt, dass es die Konzentration von zytoplasmatischem und nukleärem p53 reduziert. Sensibilisiert die Brustkrebszelllinien (BCAP37, MCF-7 und BT549) für Apoptose unter Bedingungen des Nährstoffmangels. Außerdem hemmt es nachweislich den Etoposid-induzierten Zelltod in Bax/Bak-Doppelknockout-Zellen (DKO). Normale Zelllinien (z. B. MDCK-Zellen) durchlaufen in Gegenwart von Spautin-1 keine Autophagie.
