Beschreibung

General description

Ein zellpermeables p-Imidobenzamidchinolin, endo-Diastereomer, das nachweislich die Aktivität von TNKS1/PARP5a und TNKS2/PARP5b in In-vitro-Auto-PARsylierungsassays hemmt (IC₅₀ = 131 bzw. 56 nM) und effektiv die Wnt-stimulierte Transkriptionsaktivität in L-Wnt-STF-basierten Reporter-Assays (IC₅₀ = 180 nM) unterdrückt, während es nur wenig Aktivität gegen PARP1 oder PARP2 zeigt (IC₅₀ >18,75 µM). Sowohl IWR-1-endo als auch XAV939 wirken zwar als reversible Inhibitoren des Wnt-Signalwegs und zeigen sowohl in vitro als auch in vivo ähnliche pharmakologische Wirkungen, IWR-1-endo übt seine Wirkung jedoch über eine Interaktion mit Axin aus, während XAV939 direkt an TNKS bindet.
