Beschreibung

General description

Ein hochsensitives Fluoreszenzresonanz-Energietransfer (FRET)-Peptidsubstrat (Exzitationsmaximum: ~350 nm); Emissionsmaximum: ~490), basierend auf der von der β;-Sekretasestelle der schwedischen Mutation des Amyloidpräkursorproteins abgeleiteten GVNLDAEF-Sequenz. Eine Hydrolyse des Substrats an der Leu-Asp-Bindung mittels β;-Sekretase führt zu einer verstärkten Fluoreszenz. Berichtete K_cat/k_m-Werte (min^-1/µ;M) für Pro-Memapsin 2 und seine proteolytisch aktivierte Spezies Leu^28p-Memapsin 2 sind jeweils 0,044 und 0,040 (bei pH 4,5, 37 °;C).
