Beschreibung

General description

Diese Verbindung ist ein zellpermeabler, selektiver und reversibler allosterischer Thiopyrimidon-MMP-9-Inhibitor, der spezifisch an die nichtkatalytische Stelle von MMP-9 an der PEX-Domäne mit einem K_d = 2,1 µ;M bindet und dadurch die MMP-9-Homodimerisierung und den Cross-Talk mit CD44 und dem EGFR-MAPK-Signalweg unterbricht. Es bindet nicht an die PEX-Domäne von MMP-2 oder MT1-MMP und verringert nachweislich die MMP-9-vermittelte Zellmigration in COS-1-Zellen bei 100 µ;M. Außerdem hemmt es die Zellmigration und -invasion von HT-1080- und MDA-MB-435-Krebszellen in einer dosisabhängigen Weise von 0,1 bis 100 µ;M, ohne die MMP-9-Expression oder proteolytische Aktivität in HT-1080-Zellen zu verändern. Darüber hinaus dämpft es sowohl das Wachstum des Primärtumors als auch die Metastasierung in vivo in einem Mausmodell ohne offensichtliche Toxizität (20 mg/kg, 6 Tage/Woche, i.v. und i.t.).
