Beschreibung

General description

Eine Lösung von H-89, Dihydrochlorid (Best.-Nr. 371963) in wasserfreiem DMSO. H-89 ist ein zellpermeabler, selektiver und potenter Inhibitor der Proteinkinase G (K_i = 48 nM). Hemmt andere Kinasen in mehrfach höheren Konzentrationen: Myosin-leichte-Ketten-Kinase (K_i = 28,3 µ;M), Ca²⁺/Calmodulin-abhängige Proteinkinase II (K_i = 29,7 µ;M), Proteinkinase C (K_i = 31,7 µ;M), Caseinkinase I (K_i = 38,3 µ;M) und Rho-Kinase II (IC₅₀ = 270 nM). Kann zur Unterscheidung zwischen den Wirkungen von PKA und cAMP-regulierten Guanin-Nukleotid-Austauschfaktoren (GEF), wie GEFI oder Epac (direkt durch cAMP aktiviertes Austauschprotein) und GEFII, verwendet werden. Induziert Berichten zufolge das Neuritenwachstum in NG-108-15-Zellen (~1 µ;M) durch Blockierung der Wirkung von Rho-Kinase II.
