General description
Eine zellpermeable Oxazol-Verbindung, die als selektiver CRT-Inhibitor (β-catenin responsive transcription) dient (IC50 = 8,2 nmol/l gegen die Wnt3a-abhängige SuperTopFlash STF16-luc-Reporteraktivität in HEK293-Kulturen), indem sie durch direkte β-Catenin(β-cat)-Bindung die β-cat-TCF4-Wechselwirkung stört, aber eine deutlich geringere Aktivität gegen Hedgehog, Jak/Stat oder Notch-Signalweg-abhängige Transkriptionsaktivierung aufweist. Hemmt die zelluläre Expression von CRT-Zielgenen, einschließlich WISP1, AXN2 und CYCD1, auf dosisabhängige Weise wirksam (25 bis 75 µmol/l) und hemmt die Proliferation von HCT116 (75 µmol/l), HT29 (75 µmol/l) und verschiedenen primären humanen Darmkrebskulturen (6,25 bis 100 µg/ml; mittlere IC50 ~36 µg/ml) selektiv, die für das Überleben eine anhaltende Wnt/CRT-Aktivität erfordern,.
