Beschreibung

General description

Ein zellpermeables und Blut-Hirn-Schranke-permeables Brom-zu-Chlor-substituiertes Analogon von HA14-1, das Akt durch eine spezifische Wechselwirkung mit der PtdIns(3,4,5)P3-(PIP3)-Bindungstasche der Akt-PH-Domäne-in eine Konformation bringt, die für eine Phosphorylierung durch vorgelagerte Kinasen anfällig ist. Verstärkt sowohl die basale als auch Rezeptor-vermittelte Akt-Phosphorylierung (Thr308 und Ser473) auf zeit- und dosisabhängige Weise (2 bis 4 µg/ml) bei gleichzeitiger Hemmung der Akt-Membrantranslokation in verschiedenen Zellkulturen. Verringert die durch Glutamat eingeleitete Neurotoxizität in sowohl primären Neuronenkulturen (EC₅₀ = 4 µg/ml) in vitro als auch einem murinen MCAO-Modell (Okklusion der mittleren Zerebralarterie) (40 mg/kg i.p.) in vivo effizient.
