Application
αα-Chymotrypsin von Sigma wurde zur Bestimmung der Kristallstruktur von zwei homologen Inhibitoren (pars intercerebralis-Hauptpeptid-C und pars intercerebralis-Hauptpeptid-D2v) aus dem Insekt Locusta migratoria durch die Bildung eines Komplexes mit dem Enzym eingesetzt.[1]
Biochem/physiol Actions
αα-Chymotrypsin ist eine Serinpeptidase und weist 241 Aminosäurereste in drei Polypeptidketten auf (A-Kette: 13 Reste, B-Kette: 131 Reste und C-Kette: 97 Reste), die durch Disulfidbrücken verbunden sind. Die Molekülmasse dieses Enzyms beträgt 25 kDa. Sein pH-Wert liegt bei 8,75. Es hydrolysiert Peptidbindungen an der C-terminalen Seite von Tyrosin, Phenylalanin, Tryptophan und Leucin selektiv. Ca2+ aktiviert und stabilisiert das Enzym. Das Enzym wird durch Diisopropylfluorphosphat (DFP), Phenylmethansulfonylfluorid (PMSF), N-p-Tosyl-L-phenylalanin-chlormethylketon (TPCK), Chymostatin, Aprotinin α1-Antitrypsin, α2-Macroglobulin, 10 mM Cu2+ und Hg2+ gehemmt.
Ein Serin-Protease, die Peptidbindungen mit aromatischen oder großen hydrophoben Seitenketten (Tyr, Trp, Phe, Met, Leu) am Carboxylende der Bindung hydrolysiert.
Preparation Note
Die Behandlung mit TLCK inaktiviert Trypsin, das in Chymotrypsin vorkommen kann, ohne die Chymotrypsinaktivität zu beeinträchtigen.
