General description

Parathyroid hormone-related protein (UniProt P12272; also known as Parathyroid hormone-like related protein, Parathyroid hormone-like hormone preproprotein, PLP, PTH-related protein, PTH-rP, PTHrP) is encoded by the PTHLH (also known as BDE2, HHM, PTHR, PTHRP) gene (Entrez Gene ID 5744) in human. Parathyroid hormone-related protein is initially translated as a 177 amino acid preproprotein with an N-terminal signal sequence (aa 1-24) and a propeptide sequence (aa 25-34), which are later removed to yield the mature protein. The mature protein is further cleaved to produce PTHrP[1-34] (aa 37-70), PTHrP[1-36] (aa 37-72), PTHrP[38-94] (aa 74-130), and PTHrP[107-139] (aa 143-175; also known as Osteostatin). PTHrP is produced in a wide variety of tissues, where it acts as a paracrine or autocrine factor to regulate cellular and organ growth, development, migration, differentiation, calcium transport, survival and apoptosis.
~16/6 kDa observed. Uncharacterized band(s) may appear in some lysates.

A major focus of breast cancer research is to understand the mechanisms responsible for disease progression and drug resistance. Toward that end, it has been found that approximately two thirds of all human breast carcinomas overexpress the Estrogen Receptor α (ERα) protein and it remains the primary pharmacological target for endocrine therapy1,2. The normal cellular function of ERα is as a transcription factor that mediates a wide variety of physiological processes, many of which are dependent upon phosphorylation of the receptor at specific amino acid residues3,4. Indeed, ERα is known to be phosphorylated at a multitude of different sites, yet how these all correlate to disease remains unclear5. Here, we interrogated multiple sites of ERα for phosphorylation status by screening an extensive panel of different breast cancer patient samples and other non-breast cancer tissue microarray (TMA) slide samples to determine their relevance to disease.
