Application
Mit Puffer, pH 7,5-8,0 auf das gewünschte Arbeitsvolumen verdünnen.
Endgültige Arbeitsverdünnungen müssen vom Endanwender bestimmt werden.
EMPFOHLENES IFA-PROTOKOLL:
Substrat-Objektträger:
1. Die Zellen abkratzen und in die Vertiefungen der Mikroskop-Objektträger geben.
2. Die Präparate an der Luft trocknen lassen.
3. In kaltem Aceton bei +4 °C 10 Minuten lang fixieren.
Färbung:
1. 15 μl verdünnten Polio-Antikörper zugeben.
2. In einer feuchten Kammer bei +37 °C 30 Minuten lang inkubieren.
3. In PBS 5 Minuten lang spülen. Vorgang mit frischer PBS wiederholen.
4. 15 μl verdünnten markierten Antikörper zugeben.
5. Die Schritte 2 und 3 wiederholen.
6. Mit gepuffertem Glycerol eindecken.
