Beschreibung

General description

Eine zellpermeable Chinolincarboxamid-Verbindung, die nachweislich das mitochondriale SIRT3 in einem Substrat hemmt, das AceCS2-kompetitiv ist (K_i = 0,56 µM; K_m = 2,44 µM), aber NAD⁺-unkompetitiv (K_i = 0,34 µM; K_m = 280 µM), hemmt. Es wird auch berichtet, dass es die zelluläre p53 Lys382-Acetylierung (effektive Konzentration = 10 µM in U2OS- und MEF-Kulturen) verringert und die p300 HAT-Aktivität (IC₅₀ = 9 µM) in vitro hemmt sowie therapeutische Vorteile in mehreren Mäuse- und Nagetiermodellen von Typ-2-Diabetes (100 mg/kg/Täglich p.o.) in vivo bietet. Ob und wie SRT1720 die SIRT1-Aktivität aktiviert, bleibt unklar. Auch als 25-mM-Lösung in DMSO verfügbar (Kat.-Nr. 530748).
