General description

Una disolución de fenol equilibrada con Tris es un líquido transparente cubierto con una capa de tampón Tris. Las disoluciones de fenol se pueden utilizar para una amplia variedad de aplicaciones de biología molecular. Los compuestos orgánicos como los fenoles tienen un anillo carbocíclico aromático con un grupo hidroxilo unido directamente a él. El nombre común del monohidroxibenceno es fenol.

Application

La disolución de fenol se ha utilizado:

  • como componente de la mezcla de reacción de digestión para preparar plásmidos sin ARNasas destinados a la traducción in vitro en un lisado de reticulocitos de conejo
  • para la extracción de ADN de hojas de roseta Arabidopsis thaliana
  • en la extracción de ADN de sangre en papel conservante por protocolo estándar de fenol-cloroformo
  • como componente del tampón de extracción para extraer ARN de la pulpa de la fruta
  • para la extracción de proteínas de las raíces
  • para el aislamiento del ADN fecal
  • para la extracción de ARN enjaulado[1]

Biochem/physiol Actions

El método de extracción fenol-cloroformo se emplea ampliamente para extraer ácidos nucleicos. El fenol no inhibe por completo la actividad ARNasa, pero su característica única lo hace beneficioso para el aislamiento de ácidos nucleicos cuando se usa en combinación con cloroformo y alcohol.

  1. What happens if I mix the two layers of phenol before pipetting? I prepared FIAC this way

    1. Mixing the two phenol layers does not damage this product. Phase separation can occur during handling. If layers mix, allow the solution to stand until a clear phase separation is re-established before pipetting. Temporary haziness or a visible interface after mixing is normal. Pipetting should occur only after full separation to avoid aqueous carryover and ensure consistent performance.   2. How long can you store phenol for, after adding the buffer?

    1. Following the addition of buffer, the solution may be stored for 6 months at 2–8 °C. For extended storage after opening, with or without the addition of buffer, the material may be stored at –20 °C, protected from light. This information is available in the product datasheet.     1. Yes, this phenol solution includes a separate bottle of equilibration buffer (Product No. B5658) to adjust the phenol phase to pH 7.9±0.2. The entire contents of the bottle of the buffer should be added to the phenol.

    1. Both package sizes are bottles that are sealed in a polyamide bag. This bag is placed, with additional absorbent material and padding, in a metal can with a pull tap. The final packaged item is placed in the shipping box that is most compatible with the order. This product is shipped at ambient temperature.

  6. What do I do to equilibrate the Phenol solution, Product P4557, to around pH 8?

    1. This phenol solution (Product No. P4557) includes a separate bottle of equilibration buffer (Product No. B5658) to adjust the phenol phase to pH 7.9±0.2. After addition of the buffer to the phenol, it may take about 6 hours (mixture can be kept at 2-8 °C) until the bottom phase (phenol) becomes clear. Some of the haziness in the phenol phase may be due to some air bubbles caused by mixing by inversion. There could be a hazy interface between the phenol and aqueous phase which remains after the separation.  The entire contents of the bottle of the buffer should be added to the phenol. If buffering less than the complete bottle, use at a ratio of 6.5 mL buffer to 100 mL phenol. For 10 mL of phenol, use 650 μL of buffer. For 1 mL of phenol, use 65 μL of buffer.
