General description

RNAlater es compatible con la mayor parte de los métodos de aislamiento del ARN, como el reactivo TRI y los kits de aislamiento de ARN total GenElute y de aislamiento de ARNm de mamíferos.

RNAlater se un reactivo acuoso, no tóxico para conservación de tejidos que impregna rápidamente el tejido para estabilizar y proteger in situ el ARN celular en muestras no congeladas. Los fragmentos de tejido se recogen y se sumergen inmediatamente en RNAlater para su conservación sin poner en peligro la calidad ni la cantidad del ARN. RNAlater elimina la necesidad de procesar de inmediato las muestras de tejido o de congelarlas en nitrógeno líquido para su procesamiento posterior. RNAlater conserva el ARN en los tejidos durante un máximo de 1 día a 37 °C, 1 semana a 25 °C y 1 mes a 4 °C. Los tejidos pueden conservarse también a largo plazo a -20 °C.

RNAlater se ha probado de manera exhaustiva en diversos tejidos de especies de vertebrados: cerebro, corazón, riñón, bazo, hígado, testículos, músculo esquelético, grasa, pulmón y timo. RNAlater es también efectivo para E. coli, Drosophila, cultivos de tejido, leucocitos y algunas plantas.
RNAlater puede utilizarse con varias aplicaciones subsiguientes, como aislamiento de ARNm y ARN total, histología e inmunocitoquímica, y es compatible con kits de aislamiento de Sigma.

Application

Biochem/physiol Actions

RNAlater es fácil de utilizar. Sólo hay que cortar las muestras de tejido que se vayan a conservar de forma que tengan menos de 0,5 cm por lo menos en una dimensión y sumergirlas en 5 volúmenes de RANlater. Órganos pequeños, como el riñón, el hígado o el bazo de rata pueden conservarse enteros en RNAlater. Cuando se esté listo para aislar el ARN, solo hay que retirar el tejido de RNAlater y procesar como si se acabara de recoger. Para la conservación de las células, resuspender las células sedimentadas en una pequeña cantidad de PBS antes de añadir 5-10 volúmenes de RNAlater. Antes de preparar el ARN, sedimentar las células y desechar el sobrenadante.

  8. Does RNAlater stop transcription upon addition or only preserves the RNA from degradation by RNAases?

    1. RNAlater™ is used to stabilize and protect RNA with immediate RNase inactivation. It works by denaturing proteins, including RNase. The use of RNAlater™ RNA Stabilization Solution for tissue storage does not affect the outcome of subsequent RNA expression analysis experiments compared to other processing methods. It is worth noting that proteins will be preserved in RNAlater™ however, the solution will denature the proteins. Therefore, the protein obtained from samples stored in it will still be suitable for applications such as western blotting or 2D gel electrophoresis, but not for applications that require native protein.
