General description

Chromolith® columns are made of monolithic silica with a characteristic bimodal pore structure.

Chromolith® FastGradient RP-18 endcapped 50-3 columns have macropores with 2 μm diameter, giving a column efficiency exceeding 80,000 plates/meter. The mesopores are 13 nm (130 Å), the surface modification is octadecylsilane with full end-capping. Chromolith® FastGradient RP-18 endcapped 50-3 columns fall under the USP L1 Classification. The column efficiency is comparable to a 5 μm particle size fully porous particulate column. Chromolith® FastGradient RP-18 endcapped 50-3 columns are perfectly suitable for LC-MS applications.
The column dimension of 50 mm length and 3 mm I.D. is the best choice for fast separations on any HPLC and UHPLC System.

The revolutionary, monolithic silica with its unique combination of macropores and mesopores enables:
  • Rapid separations at very low column back-pressure
  • High matrix tolerance, allowing time and cost savings with simplified workflows
  • Cost-savings due to extended column lifetimes
  • Connection of columns, flow gradients and the use of mobile phases with higher viscosity

The recommended Guard-columns are:
Chromolith® RP-18 endcapped 5-3 guard cartridges (3 pieces) [1.52005] with the Chromolith® Guard Cartridge Holder 5 mm x 3 mm [1.52004].
Holder and guard cartridges are also available as Guard cartridge set [1.52008].

The Chromolith® column coupler can be used to significantly increase the separation efficiency of Chromolith® columns by coupling several columns in series (up to 100,000 plates/per column). [1.51467].

Application

  • Analytical Method Development, Validation and Forced Degradation Study of Dapagliflozin by RP-HPLC: Utilizing the Kromasil® C8 HPLC column, this study demonstrated the column′s robustness in pharmaceutical applications, particularly for the detailed analysis of Dapagliflozin, a diabetes medication. The research highlights the column′s effectiveness in handling complex degradation studies essential for drug stability testing (Chaudhari et al., 2023).

  • Determination of Olmesartan in Bulk and Pharmaceutical Dosage Forms: This research outlines the use of the Kromasil® C8 column for high-throughput quantification of Olmesartan, showcasing the column′s precision and repeatability in both bulk and dosage form analyses. The method is vital for quality control in pharmaceutical manufacturing (Agrawal et al., 2023).
  • Quantification of a New Antifungal Drug Based on 1,3,4-Thiadiazole by HPLC-ESI/MS: Highlighting the Kromasil® C8 column′s high resolution and sensitivity, this study focuses on the development and validation of a method for quantifying a novel antifungal compound, essential for both pharmaceutical development and therapeutic monitoring (Sipkina et al., 2022).
