General description

La fibronectina 1 es una glucoproteína de la matriz extracelular codificada por el gen FN1. Se expresa en la plasma y en la superficie celular. El gen FN1se ha localizado en el cromosoma humano 2q35.
La FN aparece como un dímero proteico, unido por dos enlaces disulfuro en el extremo C-terminal. Dentro de la estructura de la fibronectina, los dominios están dispuestos de forma compacta cerca de los extremos amino y carboxilo terminales de las cadenas peptídicas. Además, pese a proceder de un solo gen, en humanos se han identificado hasta 20 variantes distintas de este gen.

Application

La fibronectina del plasma humano se ha utilizado en la determinación de la tasa de consumo de H2O2.[1] También se ha utilizado en el recubrimiento de los modelos Sylgard® para cultivo celular. También se ha utilizado en la adhesión celular y para modelar directamente la proteína sobre la película polimérica.

Biochem/physiol Actions

La función de la fibronectina abarca una amplia variedad de procesos biológicos, como la adhesión celular, el crecimiento, la migración, la cicatrización de heridas, la coagulación sanguínea y la formación de metástasis. La proteína fibronectina desempeña un papel importante en la fijación y la propagación celulares, en el control del citoesqueleto, la morfología y la diferenciación celulares. La FN1 interviene también en la formación de la matriz extracelular, la hemostasis y la trombosis. Mutaciones en la FN1 producen glomerulopatías.[2]

Preparation Note

Este producto es una disolución al 0,1% de fibronectina. La homogeneidad de la proteína se evalúa mediante inmunoelectroforesis, se analiza la presencia de endotoxinas y se ha demostrado que el material original era negativo para el VIH, la hepatitis B y la hepatitis C.

En el recubrimiento de las superficies de cultivo, la fibronectina debe diluirse en disolución salina estéril equilibrada y recubrirse con un volumen mínimo. Luego hay que dejar secar la superficie al aire durante 45 minutos a temperatura ambiente y puede conservarse durante 2-4 semanas a 2-8°C

Analysis Note

La homogeneidad proteica se evalúa mediante inmunoelectroforesis.

  1. Could Fibronectin (F0895) be used as a positive control for fibronectin in western blot assay? If so, do one need to consider any extra steps in the preparation of such ctrl sample?

    1. This product is intended for use as an attachment substrate for cell culture and has not been tested in western blotting as a positive control. However, the purification process of the starting material includes an affinity step and immunoelectrophoresis performed during Quality testing is against Anti-Fibronectin antibody F3648. While the end user would have to determine suitability, this material is expected to be suitable for use as a positive control in western blotting.

  2. How many µg/cm² would the coating concentration be if I applied a coating with a 0.1% concentration using the F0895 product?

However, the solution cannot be reused to coat culture ware surfaces multiple times; the recommended process involves coating surfaces with a minimal volume, allowing them to air dry for 45 minutes at room temperature, and then storing the cultureware at 4°C for 2-4 weeks.

    Why do you not have a protocol for using this expensive reagent?

    You have no protocol online for using this very expensive item......why? I have been waiting to chat for a total of 15 minutes to get what you should have put online anyway

    No issues, seems to work for hiPSC differentiation protocols.

    Used multiple times and different lots for hiPSC differentiation into iMeLCs. No problems with it.
