General description

Actin is a highly abundant and conserved protein. It can transition between the monomeric (G-actin) and filamentous (F-actin) forms, which is influenced by external factors. Actin is expressed in smooth, skeletal and cardiac muscles.

Application

Actin from bovine muscle has been used as an internal standard and positive control during western blotting.

Biochem/physiol Actions

Actin is involved in a wide variety of cellular processes like cell migration, phagocytosis, cell polarity and shape. It associates with myosin and aids in muscle contraction. The protein acts as a building block of microfilaments.

  1. Can you provide the protocol to polymerize the Actin from Bovine muscle?

    1. In the absence of salts, actin exists as a globular monomeric protein (G-actin). In the presence of ATP, a divalent cation (Ca2+ or Mg2+) and high salts (KCl, NaCl or MgCl2), the G-actin polymerizes to form a fibrous or filamentous actin (F-actin) which incorporates the cation and ATP (3,4). Dialysis of the F-actin solution (viscous) against water reverses the polymerization (2). The actin as it is supplied in product A3653 has been extensively dialyzed to be converted into G-Actin.
