General description

Kallikrein-1 (EC:3.4.21.35; UniProt P06870; also known as Glandular kallikrein 1, hK1, Kallikrein serine protease 1, Kidney/pancreas/salivary gland kallikrein, Renal/pancreas/salivary kallikrein 1, Tissue kallikrein) is encoded by the KLK1 (also known as KLKR) gene (Gene ID 3816) in human. Kallikrein-1 is a member of the kallikrein subfamily of serine proteases, members of which are involved in diverse physiological functions and many are used clinically as biomarkers for cancer. Kallikrein-1 is initially produced as a preproprotein with a signal (aa 1-18) and a propeptide (aa 19-24), cleavage of which releases the mature protein (aa 25-262). Kallikrein-1 is the major kallikrein expressed in the kidney, where it generates kinins (e.g. lysyl-bradykinin and bradykinin) and plays role in kidney function and blood pressure regulation through vasodilatory and natriuretic effects. Decreased kidney Kallikrein-1 level is reported in experimental settings of hypertension, diabetic nephropathy, high salt- and aminoglycoside-induced kidney injury.

GST-tagged recombinant protein corresponding to the C-terminal half of human Kallikrein-1/KLK1.

Application

Immunohistochemistry Analysis: A 1:50 dilution from a representative lot detected Kallikrein-1/KLK1 in human pancreas and human colon tissue.
This Anti-Kallikrein-1/KLK1 Antibody, clone 11C7.1 is validated for use in Western Blotting, Immunohistochemistry (Paraffin) for the detection of Kallikrein-1/KLK1.

A major focus of breast cancer research is to understand the mechanisms responsible for disease progression and drug resistance. Toward that end, it has been found that approximately two thirds of all human breast carcinomas overexpress the Estrogen Receptor α (ERα) protein and it remains the primary pharmacological target for endocrine therapy1,2. The normal cellular function of ERα is as a transcription factor that mediates a wide variety of physiological processes, many of which are dependent upon phosphorylation of the receptor at specific amino acid residues3,4. Indeed, ERα is known to be phosphorylated at a multitude of different sites, yet how these all correlate to disease remains unclear5. Here, we interrogated multiple sites of ERα for phosphorylation status by screening an extensive panel of different breast cancer patient samples and other non-breast cancer tissue microarray (TMA) slide samples to determine their relevance to disease.
