The milligram quantity of this product will vary from lot to lot. The vial content is dependent upon the protein content and enzymatic activity. The minimum protein content will be 70% and the minimum activity will be 2 units per milligram protein. As an example, a batch that reports a protein content of 100% and an activity of 2 units per milligram protein will have a vial content of 2.5 mg for the 5 unit package size. General description
La enzima conversora de angiotensina (ACE) es una dipeptidil-carboxipeptidasa que existe en las formas somática y testicular con el motivo de unión a cinc HEXXH en su centro activo. La ACE regula la presión arterial a través del sistema renina-angiotensina. La ACE eleva la presión arterial al convertir la angiotensina I en un vasoconstrictor fundamental, la angiotensina II, e inhibir un potente vasodilatador, la bradiquinina. La inhibición de la ACE es una estrategia terapéutica para la hipertensión. Algunos péptidos sintéticos inhibidores de la ACE en la clínica son el captoprilo, el enalaprilo y el lisinoprilo. En la actualidad se están desarrollando péptidos inhibidores de fuentes alimenticias naturales, o de compuestos fenólicos de origen vegetal para inhibir la ACE. La ACE desempeña un papel crucial en la fertilización al liberar las proteínas ancladas al glucosilfosfatidilinositol (GPI) en la membrana del esperma.
Application
La enzima conversora de angiotensina procedente de pulmón de conejo se ha utilizado:
- para medir el efecto inhibidor de los hidrolizados de proteína de clara de huevo sobre la actividad de la ACE mediante cromatografía líquida de alta resolución (HPLC)
- para medir la inhibición de la ACE por el pericarpio de lichi y pechuga de pollo cocida utilizando hipuril-L-histidil-L-leucina (HHL) como sustrato para la HPLC de fase inversa ((RP-HPLC)3 y la HPLC respectivamente
- en la liberación de las proteínas ancladas al GPI in vitro en unas pocas líneas celulares como HeLa y HEK293 e in vivo en el esperma de ratón.

Elimina los péptidos C-terminales de los sustratos predispuestos, por ejemplo la angiotensina I y la bradiquinina.

Idrapril, the prototype of a new class of angiotensin converting enzyme (ACE) inhibitors, competitively inhibits, with nanomolar apparent Ki, the hydrolysis of hippuryl-glycyl-glycine by rabbit lung ACE. Kinetic Ki and kon and koff values were 470 pM, 3.0±1.5×106 M-1sec-1 and 1.4±0.3×10-3 sec-1, respectively. Reference: Biochemical Pharmacology, 45(6), 1358-1362 (1993).
