General description

The silent information regulator (SIR) genes (sirtuins) are a highly conserved family of proteins, which contain one or more sirtuins present in various species from bacteria to mammals. There are seven sirtuin genes that encode seven sirtuin enzymes, SIRT1 through SIRT7, all of which are dependent on oxidized nicotinamide adenine dinucleotide (NAD+). These proteins serve as mono-ADP-ribosyltransferases and deacetylate histones as a means of silencing specific chromatin regions. Further research is still required to elicidate the link between its effects on cellular metabolism, stress resistance, and cellular proliferation and its proposed role in mediating general lifespan. It is reported that Sirt7 expression demonstrates an anti-proliferative role with tumorigenic potential, in several murine cell lines.

Specificity

This antibody recognizes the N-terminus of the expressed protein of SIRT7.

Immunogen

Epitope: N-terminus.
KLH-conjugated linear peptide corresponding to the N-terminus of the expressed protein of human SIRT7.

Application

Immunohistochemistry Analysis: A 1:00 dilution from a representative lot detected the expressed protein of SIRT7 in human liver tissue lysate.

Quality

Evaluated by Western Blot in human liver tissue lysate.

Western Blot Analysis: 0.5 µg/mL of this antibody detected the expressed protein of SIRT7 in 10 µg of human liver tissue lysate.

Target description

~45 kDa observed. An additional isoform at ~20 kDa is known to exist. An uncharacterized band at ~30 kDa has been observed in some lysates.
