General description
3-Isobutyl-1-methylxanthin/IBMX ist ein nicht selektiver, unspezifischer Inhibitor der cAMP- und cGMP-Phosphordiesterasen. IBMX dient der Auslösung von Melanogenese und kann daher als Positivkontrolle in der Melanogenese-Forschung eingesetzt werden. In der Oozytenforschung hilft IBMX bei der Aufrechterhaltung der Keimbläschen-Blockierung von Prophase-I-Oozyten.
Application
3-Isobutyl-1-methylxanthin wird wie folgt eingesetzt:
- zur Differenzierung von mesenchymalen Stammzellen (MSC)
- als Kulturmedienzusatz zur Auslösung adipogener Differenzierung
- als ein Zusatz in Krebs-Ringer-Pufferlösung mit HEPES (KRBH) zur Auslösung glucosestimulierter Insulinsekretion (GSIS)
- zur Charakterisierung von aus Fettgewebe gewonnenen mesenchymalen Stammzellen
- für cyclische Adenosinmonophosphat(cAMP)-Assays
- zur Phosphodiesterase(PDE)-Hemmung
Biochem/physiol Actions
Die Steigerung des cAMP-Grades als Folge der Phosphodiesterase-Hemmung durch IBMX aktiviert PKA, was zu verminderter Proliferation, verstärkter Differenzierung und der Induktion der Apoptose führt. IBMX hemmt die Phenylephrin-induzierte Freisetzung von 5-Hydroxytryptamin aus neuroendokrinen Epithelzellen der Atemschleimhäute (IC50: 1,3 μM). IBMX dient zudem als Adenosinrezeptorantagonist. Es hat sich gezeigt, dass IBMX die Ionenkanäle in neuromuskulären Verbindungen, GH3-Zellen und glatten Gefäßmuskelzellen hemmt. IBMX stimuliert die Calciumfreisetzung aus intrazellulären Speichern in sensorischen Neuronen.

After dissolving 3-Isobutyl-1-méthylxanthine (I5879) in DMSO or in pre-warmed methanol, where should it be stored (long term or short term)?

Solutions in DMSO may be stored at -20 °C. The stability of this material in methanol has not been investigated. However, an external publication reports storage of IBMX solutions in ethanol at 0.125 mg/mL at 4 °C for at least three months. This material is soluble at a maximum concentration of 1M in DMSO. The 200 mM concentration is cited in Plouffe, B. et al., Methods Enzymol., 484, 295-328 (2010).
This concentration is sufficient for most applications. The product is tested for solubility in pre-warmed methanol at 50 mg/mL. The preferred solvent and stock concentration is dependent upon the application and requirements of the end user.
