Application

Pullulanase is a glucanase that degrades pullulan. It is commonly used for starch-debranching as well as to study polysaccharide utilization[1].

Biochem/physiol Actions

Type I pullulanases specifically hydrolyse α-1,6 linkages, while type II pullulanases are also able to hydrolyse α-1,4 linkages.

Questions

1–7 of 7 Questions
  1. What is the optimum pH and media solution for Pullulanase activity?

    1 answer
    1. Helpful?

  2. What is the meaning of NPUN/g of pullulanase enzyme E2412?

    1 answer
    1. The activity is expressed as NPUN/g, which refers to New Pullulanase Unit Novozymes (NPUN) per milligram of protein. The international unit correspondence is described as follows: 1 NPUN = 0.35 PUN (Pullulanase Unit Novozymes), where 1 PUN is defined as the amount of enzyme that, under standard conditions, releases 1 ?mol of glucose per minute.

      Helpful?

  3. I was wondering the proportion of type 1 and type2 in pullulanase?

    1 answer
    1. Product Code E2412 contains both Pullulanase 1 and 2, but the exact ratio is considered proprietary.

      Helpful?

  4. May I know how to make 10units/mL sample from the bottle?

    1 answer
    1. The activity in the batch-specific Certificate of Analysis is expressed as NPUN/g. The international unit correspondence is described as follows: 1 NPUN = 0.35 PUN, where 1 PUN is defined as the amount of enzyme that releases 1 μmol of glucose per minute under standard conditions.

The Certificate of Analysis also reports the density (g/mL) of the product. To calculate the activity in units per milliliter (UPUN/mL), multiply the activity (NPUN/g) by the density (g/mL). Further dilution can then be performed to achieve the desired final concentration.

      Helpful?

  5. Could you let me know if there are other substances included in this product for stabilization purposes, aside from the pullulanase enzyme? If so, could you specify what those additives are?

    1 answer
    1. The composition of this product is considered proprietary by the supplier, and the components have not been provided. For inquiries about the composition, please contact the supplier directly.

      Helpful?

  6. I was wondering if product E2412-50ML is a type 1 or type 2 pullulanase (degrades alpha1,6 linkages only or both alpha1,4 and 1,6?)

    1 answer
    1. E2412 contains both Type 1 and Type 2 Pullulanases.

      Helpful?

  7. What is the molecular weight of this Pullulanase enzyme? And is there any glucose in the enzyme solution?

    1 answer
    1. The molecular weight has not been determined, however, a search of the literature indicates that the general molecular weight of Pullulanase, CAS 9075-68-7, ranges from 70 - 115 kDa. This is a product of Novozymes Corp. The manufacturer considers the formulation of this product proprietary but has noted that this material is pure Oligo-1,6-Glucosidase.
