General description

Ciprofloxacin is a second-generation broad-spectrum fluoroquinolone antibiotic with extensive antimicrobial and pharmacokinetic properties that is widely used to combat bacterial infections. Its chemical name is 1-cyclopropyl-6-fluoro-4-oxo-7-(piperazin-1-yl)-1,4-dihydroquinoline-3-carboxylic acid.[1]

Application

Ciprofloxacin is widely used in clinical practices against Gram-positive and Gram-negative bacteria and the treatment of a broad range of infections, including those of the skin, eyes, urinary tract, lower respiratory tract, gastrointestinal tract, et cetera. It can also be combined with various antimicrobial agents to combat bacterial biofilms as well as multidrug-resistant microorganisms. Its derivatives have been developed to synthesize novel antibacterials with enhanced potency and diverse antimicrobial effects including antibacterial, antifungal, anti-HIV, anti-tumor, and anti-TB properties.[1]

Biochem/physiol Actions

The mechanism of action of Ciprofloxacin is the inhibition of bacterial DNA synthesis by blocking the subunit A of DNA gyrase enzyme as well as by affecting the bacterial cell wall.

    1. This product is intended for research use only. Unfortunately, there is not a pharmaceutical grade Ciprofloxacin product currently offered. Unless otherwise stated this product is not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.

  3. Is Ciprofloxacin HCL available and is it USP grade?

    1. Product 17850 has a molecular formula of C17H18FN3O3. The form of the chemical being searched for would have a molecular formula of C17H18FN3O3.HCL,H2O. There is Ciprofloxacin HCL.H2O but only in very small quantities as a secondary pharmaceutical standard, with the catalog number PHR1044. Regarding the question about whether the products are USP grade, the answer is no. Both products are labeled as strictly for Research Use Only and, as such, are not suitable for human use.

    1. In the powder form, this product is to be stored at tightly closed at room temperature. Solutions prepared in 1% HCl at 1 mg/ml may be stored in frozen aliquots at -20 °C for up to 12 months.

    1. This product is not tested for moisture content or potency/activity. The minimum purity of this product is 98% by HPLC. A similar product, PHR1167, is tested for loss on drying, but not potency/activity. This product typically demonstrates a >99% purity as compared to the USP and EP standards.   7. If a stock solution of Cipro 17850 is solved in 0,1 HCl, at what temperature should it be stored and how long ist it stable?

    1. Unfortunately, stability studies have not been performed on this product. However, several papers have information on the solution stability of ciprofloxacin.
      (1) Stock solutions of ciprofloxacin in water at 200 mg/L, 100 mg/L, 40 mg/L, 20 mg/L and 4 mg/L can be stored at -20 °C for "at least 60 days.

    1. This product is soluble in DMSO (<1 mg/ml at 25° C), methanol, 0.1N hydrochloric acid (25 mg/ml), and ethanol (<1 mg/ml at 25° C). It is insoluble in water.

  9. What is the solubility of Product 17850, Ciprofloxacin?

    1. 17850 is the free base of ciprofloxacin. It is soluble in 0.1N HCl at 25 mg/ml.  Ciprofloxacin is poorly soluble in DMSO. For HPLC, dissolve the sample material in a mixture ofAcetonitril/Phosphoric acid pH 2.4 by ultrasonification (5 mg/10 ml solvent).

  10. Can Product 17850, Ciprofloxacin, be used to remove Mycoplasma contaminations?

    1. We have not tested the product for that application. However, there is a reference in Roche's Biochemica Newsletter Issue 1 from 1996 that describes using Ciprofloxacin in mycoplasma contamination.
