Application

Dextran labeled with fluorescein isothiocyanate for possible use in perfusion studies in animals.
FITC-dextran is used extensively in microcirculation and cell permeability research utilizing microfluorimetry. FITC-dextran has been used to study plant cell wall porosity and capillary permeability. Plasma proteins have been shown not to bind to FITC-dextran.
FITC-dextran is used in cardiovascular, microcirculation, perfusion, cell monolayer and cell membrane permeability research as fluorescent flux tracer compound that supports the measurement of processes such as blood flow, membrane damage, vascular drainage and renal elimination
Fluorescein isothiocyanate–dextran (FITC-Dextran 4) is use as a fluorescent probe to study cell permeability. FITC-Dextran 4 is used with to study permeability of epithelial cell such as intestinal and renal cells in vivo.

Questions

1–7 of 7 Questions
  1. Can FITC-dextran 4KDa be view through florescence microscope to assess the gut permeability of Balb/c mice?

    1 answer
    1. Unfortunately, there is no internal data related to using this product to assess intestinal permeability. However, FITC-Dextran 4 was previously cited in this context. Kindly review the following citations:

  2. We would like to use this Product 46944-500MG for in vivo barrier integrity assays. Is there any data on the in vivo stability in lamina propria immune cells? We do not want the FITC Dextran Assay to interfere with Flow Cytometry Analysis.

    1 answer
    1. This product is not tested for the specific application, and data on in vivo stability in lamina propria immune cells is not available. A literature search is recommended for information regarding this application.

      Helpful?

  3. Are there any fluorescent hydrophobic substitutes of similar size to FITC dextran (4,000 amu) available, considering regular dextran lacks fluorescence and is hydrophilic?

    1 answer
    1. There is only one dextran conjugated with a hydrophobic fluorescent dye. The TRITC dextran, T4037, has an average dextran weight of 4,400 amu and TRITC is slightly more hydrophobic than FITC.

      Helpful?

  4. At what excitation and emission wavelengths should Product No. 46944, Fluorescein isothiocyanate-dextran, be read?

    1 answer
    1. The excitation maximum for FITC-dextran is 490 nm, while the emission maximum is 520 nm.

      Helpful?

  5. What is the recommended storage method for stock solutions of item 46944, Fluorescein isothiocyanate-dextran?

    1 answer
    1. Once Fluorescein isothiocyanate-dextran (item 46944) is in solution, it is recommended to store it at -20ºC and utilize it within 3 months to maintain its potency. To prevent multiple freeze/thaw cycles, it is advisable to aliquot the solution.

      Helpful?

  6. After reconstitution, how can we sterilise the dextran solution?

    1 answer
    1. The FITC-Dextran solution can be sterilized by filtration.

      Helpful?

  7. What is the stability of FITC-dextran in vivo?

    1 answer
    1. These products are not tested on animals. Reports suggest a 24-hour in vivo stability.
