Application

Cycloheximide solution has been used:
  • to study its role in modulating cell death induced by tumor necrosis factor α (TNFα) in combination with the second mitochondria-derived activator of caspases (SMAC) mimetic birinapant or interferon-gamma (IFNγ), specifically under conditions that inhibit caspase-8 activity[1]
  • in the study to inhibit protein translation during the CHX-chase assays[2]
  • to treat antisense oligonucleotide (AON) transfected patient-derived fibroblasts and inhibit the nonsense-mediated decay (NMD) pathway to analyze the therapeutic potential AON and visualize the transcript containing the pseudoexon (PE)[3]

Biochem/physiol Actions

Le cycloheximide (CHX) est un antibiotique produit par S. griseus. Son activité biologique principale est l′inhibition de la traduction chez les eucaryotes, provoquant l′arrêt de la croissance et la mort cellulaire. Le CHX est couramment utilisé pour la sélection de souches de levures et de champignons résistants au CHX, pour l′inhibition contrôlée de la synthèse des protéines en vue de détecter les protéines à courte durée de vie ainsi que pour la sur-induction de l′expression de protéines, et pour l′induction de l′apoptose ou la facilitation de l′induction de l′apoptose par les récepteurs de mort cellulaire.
Tests réalisés : arrêt de la croissance cellulaire, sélection de levure résistante au cycloheximide, induction de l′apoptose, et facilitation de l′induction de l′apoptose par FasL.

Other Notes

Conserver le récipient bien fermé dans un endroit sec et bien ventilé. Les récipients ouverts doivent être refermés avec précaution et maintenus en position verticale pour éviter les fuites. Hygroscopique. Classe de stockage (TRGS 510) : Matériaux dangereux non combustibles, de catégories de toxicité aiguë 1 et 2 / très toxiques.
