General description

High quality double-stranded template DNA isolated from the thymus of male and female calves.

Application

Calf thymus DNA is exceptionally useful as a substrate for DNA polymerase assays, in the amplification of very long fragments and as a carrier DNA for precipitations.
Calf thymus DNA is exceptionally useful as a substrate for DNA polymerase assays, in the amplification of very long fragments and as a carrier DNA for precipitations. It was used as a standard for DNA quantification by fluorescent assay.[1] It was used as reference to determine the DNA content in cartilage tissue digest samples.[2][3]

Biochem/physiol Actions

42% GC; Tm = 87 °C.
% GC is the percentage of G/C base pairs. Tm is the temperature at the midpoint of the thermal denaturation profile.

Preparation Note

Dissolve the DNA gently rocking overnight to a final concentration of 1mg/ml in molecular biology grade water.
Purified by anion exchange chromatography, dialyzed against a solution of 1 mM sodium chloride, 1 mM EDTA and 1 mM Trizma HCl, pH 7.5 and lyophilized at a concentration of ~2 A260 units per mL. ~20 A260 units per mg DNA.

  1. What are the storage conditions once the DNA is resuspended? Should I keep the DNA at -20ºC or -80ºC?

    1. The solution storage and stability of this product have not been established. However, the activated or nicked version of this material, product D4522, notes that Solutions of DNA have been stored successfully for several months at 4 °C, in 10 mM Tris, pH 7.5 - 8.0, with 1 mM EDTA and without a bacteriostatic agent.   2. how much is 1 unit (i.e. in ug or mg)?

    1. One unit, based on the A260 value, is approximately 50 µg. The general conversion formula is 20 units per milligram. The 0.25 UN package size will contain approximately 12.5 µg, the 1 jUN package size will contain approximately 50 µg, and the 5UN package size will contain approximately 250 µg.
