Application

Dextran labeled with fluorescein isothiocyanate for possible use in perfusion studies in animals.
Fluorescein isothiocyanate–dextran (FITC-Dextran 70) is use as a fluorescent probe to study cell processes such as cell permeability, phagocytosis and endocytosis and to study mechanisms of biomolecular delivery. FITC-Dextran 70 may be used to study the capacity, selectivity and efficiency of encapsulation and release by drug delivery vehicles.

Other Notes

Internalisation of FITC and FITC-dextran by suspension cultured plant cells; In the study of fluid pinocytosis in leukocytes[1]

Questions

1–2 of 2 Questions
  1. After reconstituting in water, what are the storage conditions?

    1 answer
    1. The stability of this material is solution has not been determined. However, aqueous solutions of 25 mg/mL or more of FITC-Dextran are expected to be stable for 1-2 weeks when stored refrigerated at 2-8°C and protected from light. For long-term storage, aliquot and store stock solutions at ?20°C, protected from light.

  2. Is this product fixable?

    1 answer
    1. Yes, paraformaldehyde can be used to fix cells. In normal circumstances, formaldehyde is a cross-linking fixative that is used to fix protein and DNA.

Looking at formaldehyde fixation, the following are applicable:
Formaldehyde cross-links with the amino group of the N-terminal amino acid residue and the side chains of 12:
      Arginine
      Cysteine
      Histidine
      Lysine
Depending on the peptide sequence, methylol groups, Schiff-bases, and methylene bridges were form

Please note that although the term paraformaldehyde is used, paraformaldehyde exists only as a solid. Once the bonds of the polymer are broken, solutions contain only formaldehyde. Any paraformaldehyde in the solution would simply be an undissolved precipitate at the bottom of the solution.
