Descrição geral

A insulina é um hormônio polipeptídico de duas cadeias produzido pelas células β das ilhotas pancreáticas. Seu peso molecular é de cerca de 5800 Da. As cadeias α e β são ligadas por duas pontes dissulfeto entre as cadeias. A cadeia α contém uma ponte dissulfeto dentro da cadeia.

Aplicação

A solução de insulina humana tem sido usada como suplemento no meio de Eagle modificado (DMEM) de Dulbecco para cultura de linhagem celular epitelial e linhagem celular endometrial humana, meio essencial mínimo Eagle para cultura de linhagem celular de adenocarcinoma de mama humano e meio 199 para cultivar a linhagem de células ovarianas normais.
Solução de insulina humana é recomendada para uso em aplicações de cultura celular em concentração de 0,5 a 1 ml/l por litro de meio.
A solução de insulina humana foi usada para teste de tolerância de insulina em camundongos e ratos.[1][2] Ela também foi usada para diferenciação adipogênica de culturas de crescimento derivadas de células-tronco embrionárias humanas (hESCs) e células estromais mesenquimais humanas (hMSCs).[3][4]

Ações bioquímicas/fisiológicas

A insulina desempenha um papel em vários processos celulares, como promover o crescimento celular, regular o ciclo celular e facilitar a captação de glicose. É comumente usada como suplemento proteico em meios de cultura de células de mamíferos livres de soro para ajudar a promover o crescimento ideal e a manutenção de células de mamíferos.

    1. For research applications, freezing insulin solution has been a common pratice. However, solutions available on the commercial market for therapeutic use are never frozen, in order to maintain maximum activity.
