General description

We are committed to bringing you Greener Alternative Products, which adhere to one or more of The 12 Principles of Greener Chemistry. This product has been enhanced for energy efficiency and waste prevention when used in starch ethanol research. For more information see the article in biofiles.

Application

Amyloglucosidase from Aspergillus niger is used to hydrolyze α-D-glucosides. It may be used in the brewing of beer and in the production of bread and juices. Amyloglucosidase, from Sigma, has been used to hydrolyze glycogen into glucose monomers in order to study lipid accumulation in skeletal muscle [1].

Biochem/physiol Actions

Amyloglucosidase from Aspergillus niger is capable of hydrolyzing the α-D-(1-4), the α-D-(1-6), and the α-D-(1-3) glucosidic bonds of oligosaccharides.[2] Amyloglucosidase is an extracellular enzyme that converts starch to dextrins and glucose. The enzyme is used in the starch-processing industry in the commercial production of D-glucose from corn syrup.[3]

Physical form

Lyophilized powder containing less than 0.02% glucose

Other Notes

One unit will liberate 1.0 mg of glucose from starch in 3 min at pH 4.5 at 55 °C.

It has been proposed that malto-oligosaccharides (MOSs) are possibly recycled back into amylopectin biosynthesis via the sequential reactions catalyzed by plastidial α-glucan phosphorylase 1 (Pho1) and disproportionating enzyme 1 (Dpe1). In the present study, the reciprocal co-immunoprecipitation experiments using specific

Questions

1–6 of 6 Questions
  1. How do you determine how many units there are in the sample I have, or how much substrate it will break down?

    1 answer
    1. This product is assigned an activity specification of 30 - 60 units per milligram protein. The minimum protein content is 80%. At the minimum protein content, each 5 mg vial will contain 120 - 240 units. This information is lot-specific and reported in the Certificate of Analysis. 2. What is the shelf life of a solution (1 mg/mL in water) of Product A7420, Amyloglucosidase from Aspergillus niger and how should it be stored?

    1 answer
    1. The stability of this product in solution has not been evaluated. However, 172426 Glucoamylase (syn. Amyloglucosides) from Rhizopus, notes that solutions are stable when stored frozen in aliquots at -20°C for up to 3 months. Similar stability would be expected for A7420. Avoid freeze/thaw cycles of solutions.
