Application

Insulin solution from bovine pancreas has been used:
  • to study the effect of exogenous insulin on plasma concentrations of glucose, insulin, insulin-like growth factor I (IGF-I), and progesterone (P4) in pubertal beef heifers[1]
  • to study the insulin responsiveness of the placenta during pregnancy complications using primary human trophoblast cells and placental villous explants[2]
  • to study the effect of insulin receptor signaling dendritic spine density in neonatal mouse hippocampal neurons in vitro[3]

Biochem/physiol Actions

Insulin is a major glucose-lowering hormone, insufficiency of which causes hyperglycemia observed in type 2 diabetes.[4]
Two-chain polypeptide hormone produced by the β-cells of pancreatic islets. Its molecular weight is ~5800 Da. The α and β chains are joined by two interchain disulfide bonds. The α chain contains an intrachain disulfide bond. Insulin regulates the cellular uptake, utilization, and storage of glucose, amino acids, and fatty acids and inhibits the breakdown of glycogen, protein, and fat.

    1. This stock solution may be diluted with sterile water, medium, or an aqueous buffer such as HEPES or phosphate-buffered saline. Sterile saline is also acceptable for dilution.

  2. Can I freeze the aliquots at -20 or -80 for the long-term storage?

    1. The USP recommends that solutions of insulin be stored at 2-8°C. It is not recommended to freeze solutions of insulin, as it can aggregate. Sterile solutions should be stable for at least 1 year at 2-8°C.

    1. The product has not been validated for rat cell culture application. However, based on the following citations, it appears to work well with rat cell culture:

      •Isolation, Transfection, and Long-Term Culture of Adult Mouse and Rat Cardiomyocytes
      published in J Vis Exp. 2020 Oct 10;(164):10.3791/61073. PMID: 33104067.

      •Collagen hydrogels with controllable combined cues of elasticity and topography to regulate cellular processes" published in Biomed Mater. 2021 Jun 11;16(4). PMID: 34030146.

  5. Does this solution contain zinc? How would that affect how quickly the insulin would act in a mammalian cell culture?

    1. The product contains 10 mg/mL insulin in 25 mM HEPES, pH 8.2, but it does not include zinc. Currently, the effect of zinc on insulin signaling is not characterized. However, it is noted that zinc appears to be able to enhance insulin-mediated IR tyrosine phosphorylation according to the article Biol Trace Elem Res. 2004 Oct;101(1):19-36. Effect of chromium and zinc on insulin signaling in skeletal muscle cells. PMID: 15516700.

  6. is this product suitable for 3T3-L1 differentiation?

    1. This product is supplied as a sterile bovine insulin solution at 10 mg/mL and would be suitable for diffentiation assays. This item has not been tested on 3T3-L1 cells, but is part of the differentiation protocol for the WT-1 preadipocyte cell line, product SCC255. See the link below to review this protocol:

  8. What is the comparison between International Units (IU) and USP units for insulin?

    1. The international unit (IU) of insulin is based on the International Standard Preparation of Bioassay (1958). The standard preparation is a mixture of 52% bovine and 48% porcine insulin, containing 24 units/mg. Reference: Martindale: The Extra Pharmacopoeia, 29th ed., p. 392. High purity bovine pancreatic insulin typically assays at approximately 27 USP units per mg. So the IU and USP unit are not identical, but they are approximately 1:1.

    1. This solution is made using insulin Product No. I5500.  The potency specification for the I5500 is not less than 27 USP units/mg.  However, we do not test for potency of the final product of this solution.  It is possible some activity is lost during the manufacturing process.
