Biochem/physiol Actions

β-D-Glucan is a biomarker used for the fungal infection, candidiasis. In a clinical study β-D-glucan levels were found to have possible usefulness in the prediction of patient outcomes.[1]

Questions

1–4 of 4 Questions
  1. How to fully dissolve powder-formed beta-Glucan into water to make a stock at 10-20 mg/mL?

    1 answer
    1. For the preparation of a 10 mg/mL solution, 1.0 g of β-glucan should be accurately weighed into a 120 mL dry Pyrex beaker. Add 5 mL of 95% ethanol to wet the sample. A magnetic stirrer bar should be added, followed by 90 mL of distilled water while stirring the slurry on a hot-plate magnetic stirrer. The heat should be adjusted to 120°C and the mixture stirred vigorously. The beaker should be loosely covered with aluminum foil, and stirring should continue vigorously. When the solution begins to boil, turn off the heat but continue stirring until the β-glucan completely dissolves (approximately 10 minutes). The volume of the solution should then be adjusted to 100 mL. This solution may be very slightly turbid due to the presence of trace amounts of protein. A 20 mg/mL solution may be achievable using the same process with more β-glucan, but validation has only been confirmed up to 10 mg/mL.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1 answer
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 answer
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf

  4. What is the recommended method for reconstituting G6513, β-D-Glucan derived from barley?

    1 answer
    1. The β-D-Glucan from barley (G6513) is soluble in water at a concentration of 20 mg/mL, as indicated by a test point.
