General description

Insulin is a two-chain polypeptide hormone produced by the β-cells of pancreatic islets. Its molecular weight is ~5800 Da. The α and β chains are joined by two interchain disulfide bonds. The α chain contains an intrachain disulfide bond.

Application

Insulin solution human has been used as a supplement in Dulbecco′s modified eagle′s medium (DMEM) to culture epithelial cell line and human endometrial cell line, minimum essential medium eagle to culture human breast adenocarcinoma cell line and medium 199 to culture normal ovarian cell line.
Insulin solution human is recommended for use in cell culture applications at 0.5 to 1mL per liter of medium.
Insulin solution human has been used for insulin tolerance test in mice and rats.[1] It has also been used for adipogenic differentiation of human embryonic stem cells (hESCs)-derived outgrowth cultures and hMSCs (human mesenchymal stromal cells).[2]

Biochem/physiol Actions

Insulin plays a role in various cellular processes, such as promoting cell growth, regulating the cell cycle, and facilitating glucose uptake. It is commonly used as a protein supplement in serum-free mammalian cell culture media to help promote the optimal growth and maintenance of mammalian cells.

OCT4 (Octamer-binding transcription factor 4) is essential for embryonic stem cell self-renewal. Here we show that OCT4 increases the aggressiveness of high-grade serous ovarian cancer (HG-SOC) by inactivating the Retinoblastoma tumor suppressor pathway and enhancing mitotic stability in cancer cells.

Questions

1–10 of 22 Questions
  1. some vendors provide insulin solution in low ph, how come this one in neutral ph

    1 answer
    1. This product is formulated at a neutral to slightly alkaline pH using a HEPES buffer (pH 7.9–8.5) to enhance long-term stability and solubility during storage. This approach differs from some vendors who use a low pH formulation for immediate solubilization.

      Helpful?

  2. In which solution was insulin dissolved?

    1 answer
    1. This is in HEPES.  10mg/mL concentration in 25 mM Hepes

      Helpful?

  3. What's the half-life of this insulin within the media at 37C?

    1 answer
    1. Unfortunately, the half-life of this insulin within the media at 37 °C has not been tested.

      Helpful?

  4. For how long this can be stored @ RT?

    1 answer
    1. This item is stored refrigerated at 2-8°C. However, the product is shipped at ambient temperature. Shipping routes are configured to minimize transit time.
Refrigerated products that ship at ambient temperature are considered stable up to 2 weeks for transit time.

      Helpful?

  5. Can I aliquot the insulin and freeze it? Is it still functional after thawing?

    1 answer
    1. This product is recommended to be stored at 2-8°C. For research applications, freezing insulin solution is a common practice. However, solutions available on the commercial market for therapeutic use are not frozen to maintain maximum activity. No stability studies have been performed on the shelf life of frozen insulin solutions.

      Helpful?

  6. What is the concentration of this solution in (Units/mL) i.e. if my stock is at a concentration of 11.1 mg/mL, what is that in Units/mL?

    1 answer
    1. The specification for the product does not include units of potency.
However, the potency is typically about 28 -29 IU/mg.

      Helpful?

  7. What is the source of this insulin and is the solutuion expected to also contain proinsulin?

    1 answer
    1. This product is a biosynthetic product, produced by recombinant expression in yeast (Saccharomyces cerevisiae), with a synthetic gene insert encoding human insulin precursor. The proinsulin content is not among the testing specifications for this product.

      Helpful?

  8. How is this insulin produced? In E.Coli? Yeast?...?

    1 answer
    1. This product is prepared from recombinant human insulin produced in Saccharomyces cerevisiae.

      Helpful?

  9. Is this product suitable for insulin tolerance tests in mice

    1 answer
    1. This item has not been specifically tested for suitability in insulin tolerance. However, the product has been used successfully in a number of published papers. Please see the link below to review one such article:
