General description

Heparan sulfate proteoglycan (HSPG), a glycoprotein, is an integral part of the basement membrane. It is composed of a core protein covalently bound to heparan sulfate chains. There are three groups of HSPGs: membrane HSPGs, secreted extracellular matrix HSPGs, and the secretory vesicle proteoglycan.

Application

Heparan sulfate proteoglycan has been used:

  • in phosphate buffered saline (PBS) and added to fresh citrated blood samples to achieve trauma-equivalent concentrations in a research study
  • solid phase binding assay[1]
  • for fabrication of extracellular matrix (ECM) microarray slides

Biochem/physiol Actions

Heparan sulfate proteoglycans (HSPGs) have multiple functions in various biological processes. They contribute to basement membrane structure and provide a matrix for cell migration. HSPGs in secretory vesicles, such as serglycin, play a role in packaging granular contents, maintaining proteases activity, and regulating biological activities after secretion. HSPGs can bind and protect cytokines, growth factors, and chemokines, facilitating the formation of gradients essential for cell specification and leukocyte recruitment. They act as receptors for proteases and protease inhibitors, regulate cell-ECM attachment, cell-cell interactions, and cell motility, and can cooperate with growth factor receptors. HSPGs also function as endocytic receptors for ligand clearance and are involved in lipoprotein metabolism and morphogen gradient formation. It also acts as a docking molecule for matrilysin and other matrix metalloproteinases. Heparan Sulfate promotes the attachment of cells in culture and induces high-affinity binding of FGF-basic to cells deficient in heparin sulfate and soluble FGF receptors at a concentration of 10-100 ng/mL.

Preparation Note

This product is isolated from the basement membrane of Engelbreth-Holm-Swarm mouse sarcoma in a solution containing 50 mM Tris HCl at pH 7.4, 150 mM NaCl, 1 mM EDTA, and 0.1 mM PMSF.

Other Notes

Heparan sulfate proteoglycan is a large biomolecule composed of a core protein covalently bound to heparin sulfate chains that is an integral part of the basement membrane. The number of polysaccharide chains and the size of the core protein are variable according to the source.
