Gene Information

General description

C1q, together with C1r and C1s, in the ratio of 1:2:2, form the C1 complex which is the first component of the classical complement pathway. C1q is composed of 18 polypeptide chains (six A, six B, six C) (MW 460 kDa). All contain an 81-aa collagen-like region composed of (Gly-Xaa-Yaa) repeating sequences close to the N-terminus. Three chains (A1B1C1) form a triple helix with the C-terminus forming the globular heads which may be structurally and functionally distinct domains.

Application

Complement component C1q is an important regulator factor for platelet activation. This has been a topic for research, as platelet-leukocyte aggregates play an important role in inflammatory conditions such as coronary heart disease. In particular, C1q has been shown to inhibit collagen induced aggregation and enhance production of reactive oxygen species (ROS).

Biochem/physiol Actions

C1q deficiency, either because of rare homozygous genetic defect, or transcriptional/translational defects, is almost certainly a cause of systemic lupus erythematosus (SLE). The three C1q subunits are expressed through a novel mechanism of transcriptional synchronization.

  1. The product is given at a concentration of approx. 1mg/ml. The product specification sheet says that product solubilized is clear, under conditions of 0.1 mL + 0.2 mL H2O. Is the concentration 1 mg/ml related to a volume of 0.1 mL (or 0.3 mL; or other?)?

    1. The complement is supplied in solution at a concentration of approximately 1 mg/mL, with the lot-specific certificate of analysis providing the exact concentration. Solubility is tested by diluting 0.1 mL of the complement stock solution with 0.2 mL of H2O, resulting in a final volume of 0.3 mL. This dilution ratio yields a complement concentration of approximately 0.33 mg/mL.

  2. I ordered the C1740-1MG with lot#0000356664. I would like to know the protein sequences and the extinction coefficient about this protein?

    1. This product corresponds to purified 'native' human C1q. Its sequence is not determined in-house. Please refer to published sequences, e.g. in GenBank for UniProt accession nos. P02745, P02746, & P02747.

  3. what is the size of the protein in WB ? when I test it's size it was approximately 24 KD ( the same size as c1q-c )

    1. C1q is composed of 18 polypeptide chains (six A, six B, six C). When prepared in denaturing sample buffer, and run on SDS PAGE, the chains will likely dissociate and run at their individual molecular weights. The Uniprot entries for C1qA, C1qB, and C1qC list masses of 26,017, 26,722, and 25,774, respectively.
