Giemsa solution should never be diluted in PBS. Giemsa solution is typically diluted with either deionized water or a low molarity phosphate buffer. Weise and Sorsensen phosphate buffer solutions are typically used. Dilutions can range from 1:10 to 1:80. The most common range for dilutions are either 1:20 or 1:40.
