General description

Extract-N-Amp PCR ReadyMix is a specially formulated hot start PCR master mix for direct PCR amplification from the extract.
PCR ReadyMix is intended for use with Sigma′s Extract-N-Amp Plant PCR kit and Extract-N-Amp Tissue PCR Kit. All Extract-N-Amp kits include a PCR ReadyMix sufficient for one PCR reaction per extraction. However, if additional PCR reactions are required, supplemental PCR ReadyMix may be needed.

Application

Extract-N-Amp PCR ReadyMix has been used for the following applications:
  • Genomic DNA extraction[1]
  • Fungal DNA extraction and amplification by polymerase chain reaction (PCR) reactions[2]
  • Direct PCR amplification[3][4]
  • Genotyping[5][6]
  • Real-Time PCR[7][8]

Modern maize hybrids often contain biotech and native traits. To-date all biotech traits have been randomly inserted in the genome. Consequently, developing hybrids with multiple traits is expensive, time-consuming, and complex. Here we report using CRISPR-Cas9 to generate a complex

In Dutch laboratories molecular detection of B. pertussis and B. parapertussis is commonly based on insertion sequences IS481 and IS1001, respectively. Both IS elements are more widely spread among Bordetella species. Both Bordetella holmesii, and B. bronchiseptica can harbour IS481.

Questions

1–3 of 3 Questions
  1. Sehr geehrte Damen und Herren, sollen wir nach dem PCR Lauf, Ladepuffer dazugeben?

    1 answer
    1. The amplified DNA should be loaded onto an agarose gel after the PCR is completed by adding a separate loading buffer or tracking dye, such as Gel Loading Solution, Catalog Number G2526.

      Helpful?

  2. Can I use the TriTrack DNA Loading Dye or the TrakitCyanoOrange Loading Buffer with the ReadyMix??

    1 answer
    1. The choice of loading dye will not impact the performance the the E3004 - Extract-N-Amp™ PCR ReadyMix. While we have not tested loading dyes outside of the product, G2526, any commercial DNA loading dye is expected to be compatible.

      Helpful?

  3. What is the polymerase used for this kit? I am trying to calculate Tm for my PCR, but I would need to know what type of polymerase this kit uses. I am guessing something close to Q5 Hotstart?

    1 answer
    1. This kit employs Taq DNA polymerase (Product D6677).
