Application

The kit is used for the detection of DHFR activity and screening of DHFR inhibitors. The assay is based on the ability of DHFR to catalyze the reversible NADPH-dependent reduction of dihydrofolic acid to tetrahydrofolic acid. The reaction progress is followed by monitoring the decrease in absorbance at 340 nm.

Biochem/physiol Actions

DHFR (dihydrofolate reductase) is an enzyme that catalyzes a reaction essential for the biosynthesis of nucleotidic bases of DNA. Blocking the enzyme causes cell death as a result of DNA synthesis inhibition. DHFR is an excellent target for anti-tumor drugs.

  1. 1. what is the origin of this DHFR. Is it from Human, E. coli or any other. 2. I want to relate antibacterial activity of my compound with this enzyme assay kit. Is it obvious.

    1. The Dihydrofolate Reductase that is provided in the kit is product D6566, a recombinant human enzyme that is expressed in E. coli. Please see the link below to review the datasheet for this component:

  2. I want to measure activity in protein variants that have different cellular concentration (protein stability), will this affect my activity?

    1. It is unclear the relation between cellular concentration and protein stability. It seems the intention is to produce dihydrofolate reductase recombinantly and then measure the activity. Protein stability is dependent on several different factors such as pH, temperature, buffering conditions, storage, purification, etc. If a protein is not stable it can certainly affect the activity. Testing would be necessary to determine the overall impact on the activity as you optimize your process.
