General description

IgG antibody subtype is the most abundant serum immunoglobulins of the immune system. It is secreted by B cells and is found in blood and extracellular fluids and provides protection from infections caused by bacteria, fungi and viruses. Maternal IgG is transferred to fetus through the placenta that is vital for immune defence of the neonate against infections[1]
Rabbit IgG is purified from normal rabbit serum by fractionation and ion-exchange chromatography.

Application

Purified rabbit IgG may be used as a reference antigen, standard, blocking agent, or coating protein in a variety of immunoassays including ELISA, dot immunobinding, Western immunoblotting, immunodiffusion, and immunoelectrophoresis. Other applications include starting materials for the preparation of immunogens and solid phase immunoadsorbents. Rabbit IgG was used to reduce non-specific binding in immunoprecipitation of protein extracts[2][3] and as control antibody in radioimmunoassay[4] immunocytochemistry[2] and immunohistochemistry.[5] Rabbit IgG was also used as a standard to demonstrate the performance of land-contrast (LC) BioCD (protein array) and in neutralization assays.[6]

  1. How can I quantify how much protein I reconstituted if the specification sheet doesn’t report an extinction coefficient for the anti-IgG?

    1. The molar extinction coefficient of rabbit IgG at 280 nm is approximately 210,000 M⁻¹cm⁻¹. The datasheet states that the protein concentration should be determined after reconstitution by measuring the absorbance at 280 nm, with an extinction coefficient of a 1% solution = 14.

      It is also necessary to note that this product is normal rabbit IgG. It is not anti-IgG

  4. How does the storage temperature relate to shipping conditions?

    1. The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.
