General description

Toluidine blue (C.l. 52040) is a diaminothiazine dye.It is also known as TBO, tolonium chloride, and toluidine blue O.

Application

Toluidine Blue is suitable for use in the following applications:
  1. to demonstrate amyloid; DNA in plant material, mast cells, Nissl substance, nuclei, and sulfatides
  2. in metachromatic staining of sulfated mucins and related materials
  3. as staining agent to identify the development and accumulation of the cartilage matrix, proteoglycans for isolation and culture of mesenchymal stem cells from mouse compact bone.
  4. staining of mucins in water-miscible resins and of nucleic acids in epoxy resins(1)

Biochem/physiol Actions

Toluidine Blue is a basic thiazine metachromatic dye, which is used for selectively staining acidic tissue components such as carboxylates, sulfates, and phosphate radicals.It is found to have an affinity for nucleic acids and therefore binds to the nuclear material of tissues with a high DNA and RNA content.

    1. The determination of the pH of Toluidine Blue powder in distilled water is not a quality specification. Section 9 of the Safety Data Sheet states the pH data is not available. It is reported elsewhere in the literature that the powder has a pH of between 2 and 2.5, but this information has not been confirmed for product 89640. The literature also reports the pKA for toluidine blue is around pH 4.5, but there are other pKa values reported in the literature. The pH of distilled water is typically around 4.2. Toluidine Blue has various uses. Some procedures call for dissolving the Toluidine Blue in water, but procedures that recommend diluting in various alcohol concentrations are also common.
