SYBR Green qPCR is utilized to determine the relative silencing effect of specific shRNAs. The term "validated" refers to shRNA sequences that have been tested and confirmed effective in silencing. This validation involves testing in specific cell lines and measuring the mean knockdown level through lentiviral transduction. The evaluation of knockdown is conducted by measuring mRNA transcript levels post-lentiviral transduction with SYBR Green qPCR and comparing these levels to those achieved with the MISSION pLKO.1-puro Control Transduction Particles (SHC001V). For instance, a mean knockdown level of 0.90 indicates a 90% reduction at the mRNA level. Typically, validation does not extend to the protein level, as the primary target of shRNA is mRNA expression, which subsequently influences protein expression.

General description
Our comprehensive shRNA product offering consists of over 150,000 pre-cloned shRNA constructs targeting more than 15,000 human and 15,000 mouse genes. MISSION® TRC shRNA purified plasmid DNA format takes the hassle out of preparing DNA from each construct. DNA may be used directly for transient or stable transfection. Additionally these plasmids can be combined with packaging plasmids in a packaging cell line to produce lentiviral particles. 1 μg of DNA is provided for each construct in the target set.
Application
MISSION® shRNA has been used:
- in the generation of ZO-1 (zonula occludens-1) knockdown
- to knock down LIF (leukemia inhibitory factor) expression in isogenic MCF10 model
- to knock down nuclear factor erythroid 2 like 1, in n MIN6 pancreatic β-cells
