General description

The EmbryoMax ES Cell Qualified FBS is exclusively of US origin. Each lot is sterile filtered and mycoplasma, endotoxin, and virus tested. Additionally, each lot is prescreened for plating efficiency and maintenance of undifferentiated morphology of mouse embryonic stem cells. A Certificate of Analysis is available upon request for each lot of EmbryoMax ES Cell Qualified FBS.
Note: Precipitates may develop upon freezing and/or thawing of this product. If precipitates are present, filter product through a 0.2 micron filter. This occurrence does not impact product performance.
The maintanence, expansion, and assay of many stem cell and precursor cell cultures relies on media rich in a variety of factors, often supplied by fetal bovine serum (FBS).

"As transgenic mouse core laboratories know well, mouse models are a powerful means of determining the function of mammalian genes in the context of whole organisms—especially since over 95% of the mouse genome is similar to the human genome. However, you may not know that: 1. Laboratory mouse strains are descended from ancestors in modern-day Pakistan. 2. The first inbred strain of lab mouse was created in 1909. Male mice, when treated with anticancer drugs, have been shown to pass on DNA instability to their offspring. 5. The founder of Jackson Laboratories, C. C. Little, once had lunch with Walt Disney to request that an animated film be created linking Mickey Mouse and the laboratory mouse. The request was denied."

In order to create a quantitatively defined diffusion gradient that is stable enough for long-term, live cell imaging over the course of days, we developed a microfluidic gradient plate. This plate, designed for use on the CellASIC™ ONIX Microfluidic Platform, enables precision-controlled chemoattractant diffusion across perfusion barriers to create a spatial gradient in the culture area. On the other end of each culture area, there are flow outlets. Together, the perfusion inlets and outlets form a continuous flow “infinite source/sink” that maintains a stable concentration gradient profile for days. The flexible format of the plate enables changes in gradient directionality, turning gradients on and off, and toggling between gradient and single solution exposure.

The cell culture protocols described in this instructional manual include the in vitro culture of murine ES cells using EmbryoMax products along with ESGRO® mLIF medium supplement, as well as feeder-free and serum-free ES cell culture using the ESGRO Complete™ line of products. Also included in this instructional manual are: methods for serum-free neuronal differentiation of mouse ES cells, iPS cell generation using STEMCCA™ lentivirus kits, cre-recombinase mediated excision of STEMCCA™ genes from iPS cells, derivation and rescue of new and existing ES cell lines using RESGRO™ Culture Medium and ESGRO Complete™ system.
