Allgemeine Beschreibung

RPMI-1640 Medium wurde 1966 im Roswell Park Memorial Institute von Moore und seinen Kollegen entwickelt. Als Modifikation des McCoy-5A-Mediums wurde es für lymphoblastoide Zellen in Suspensionskultur formuliert, hat sich aber seitdem auch für viele adhärente Zellen als geeignet erwiesen.
RPMI 1640-Medium wurde 1966 am Roswell Park Memorial Institute von Moore und seinen Kollegen entwickelt. Als eine Modifikation von McCoy′s 5A-Medium wurde es formuliert, um lymphoblastoide Zellen in Suspensionskultur zu ernähren, aber seither wurde aufgezeigt, dass es ein breites Spektrum von Zellen ernährt, die verankerungsabhängig sind. Ursprünglich war es dazu gedacht, mit einer Serumsergänzung verwendet zu werden, doch es wurde aufgezeigt, dass RPMI 1640 verschiedene Zelllinien in Abwesenheit von Serum ernährt. Es wird auch weitverbreitet in Fusionsprotokollen und beim Wachstum von Hybridzellen eingesetzt. Dieses Medium eignet sich für die Kultivierung von normalen und neoplastischen humanen Leukozyten.

Anwendung

RPMI-1640-Medium wird als Medium zur Erhaltung von Zelllinien verwendet und dient für verschiedene Zellen als Medium.[1][2]

  1. I bought this product and realized that it does not contain Hepes. Can I add hepes?

    1. This product utilizes a bicarbonate buffering system. However, there are no anticipated problems with the addition of HEPES. We do offer RPMI formulations containing both sodium bicarbonate and HEPES, such as Products R5886 & R7638.

  2. Considering that I have to use an RPMI medium containing 4mM of l-Glutamine and 10mM of non-essential amino acids, can I use the product R8748 without adding anything?

  4. Can Product R8758, RPMI-1640 Medium, be frozen?

    1. We do not recommend freezing cell culture media. A precipitate may form upon thawing.  Also, this can cause a loss of amino acids, which may yield low cell growth.

    1. This formulation of RPMI already contains L-glutamine at 0.3 g/L.  It does not require further supplementation with L-glutamine or the L-glutamine dipeptide (Product No. G8541).
