General description
Dulbeccos Phosphatgepufferte Kochsalzlösung (DPBS) ist eine ausgeglichene Salzlösung (BSS), die für die Handhabung und Kultivierung von Säugetierzellen eingesetzt wird. DPBS-Puffer wird zum Spülen, Waschen und Verdünnen von Säugetierzellen und Beibehaltung des osmotischen Gleichgewichts verwendet. Es stellt Wasser und anorganische Ionen bereit, die für den normalen Zellstoffwechsel unerlässlich sind. DPBS mit Calcium und Magnesium erleichtert die Bindung und Aggregation von Zellen. Dulbeccos PBS ohne diese Ionen kann zum Waschen und Spülen suspendierter Zellen eingesetzt werden. Der Phosphatpuffer hält den pH-Wert im physiologischen Bereich.
Application
Anwendungsgebiete von Dulbeccos Phosphatgepufferter Kochsalzlösung:
- Zum Verdünnen frischer Erythrozyten zur Untersuchung des Einfangens von Erythrozyten-Aggregaten.
- Zum Waschen der Wells einer Mikrotiterplatte.[1]
- Zum Waschen der Zellen vor dem Resuspendieren in Lysepuffer für die DNA-Isolierung.[2]
- Als Bestandteil des Embryo-Aufbewahrungsmediums zur Vitrifizierung von Embryonen.

What is a Salt Toxicity Test?

The Cell Culture assay is called a Salt Toxicity Test (previously called Cell Culture Salt Testing). A more complete description is below:Cells are harvested and counted. They are resuspended in DPBS and incubated at 37°C, 5% CO2 for 1 hour. The cells are counted and %viability determined. The cells are then cultured for an additional 7 hours (total of 8 hours). The cells are counted again and the % viability determined. The viability at 1 hr has to be >95% of the original viability. The viability at 8 hrs must be >75% of the original viability.
