General description

Poly-Prep slides are made of glass and designed for cell adhesion. The slides are coated with Poly-L-Lysine, a non-specific attachment factor for cells useful in promoting cell adhesion on solid substrates. Poly-L-lysine enhances binding between the cell membrane and the cultured surface.

Application

Poly-Prep Slides are used:
  • To mount liver and lung cell sections and mounted on poly-prep slides.
  • To prove better cell adhesion in clinical bronchoalveolar lavage fluid samples.
  • To determine pAkt (alpha serine/threonine-protein kinase) expression by immunohistochemistry.
  • In immunofluorescence imaging of nuclear translocation of nuclear factor κB(NFκB) in A549 cells.
  • To study the phospholipase C-zeta activity of sperm.
  • To prepare coelomocytes smears, which were further subjected to antibody staining.
Overall, glass slides coated with poly-L-lysine for tissue section adhesion in routine histological uses. And, it is not recommended for cell culture or microarray applications.

  4. Would you recommend wiping the slides with ethanol before use? What alternatives can be used if not ethanol? I'm seeing some artifacts in microscopy which is not from the dyes or tissue.

    1. Assuming the Poly-Prep Slides are clean, it is doubtful that dipping the slides into ethanol or other solvents will decrease the artifact you are seeing. Poly-Prep slides have been noted to have blue fluorescence in the UV range. There are reference articles that mention blue fluorescence of L-Lysine in the UV range. The full PDF can be downloaded at the URL provided below. In house experience with the polymer matches that of the monomer.

  5. What's size of the slides?

    1. These slides are rectangular with frosting at one edge.
      The dimensions are given as 3 inches (~75mm) x 1 inch (~25mm) with a thickness of 1mm.
      The coated area is approx 18 to 20 mm from one edge.

  6. How can the side of the slide that is poly-L-lysine coated be differentiated?

    1. The coating is visible as white. Using a lead pencil to identify which side of the surface has been coated is suggested, as the lead will mark the coated side but not the uncoated glass.
