Descrizione generale
I nucleotidi sono molecole organiche che fungono da monomeri, o subunità, degli acidi nucleici (come DNA e RNA). I nucleotidi, i mattoncini costitutivi degli acidi nucleici, consistono di una base azotata (purina o pirimidina), uno zucchero con cinque atomi di carbonio (ribosio o desossiribosio) e almeno un gruppo fosfato. Un nucleoside insieme con un gruppo fosfato costituisce un nucleotide. Questa miscela di desossinucleotidi è una pratica soluzione premiscelata di dNTP, contenente 10 mM ciascuno dei sali sodici di dATP, dCTP, dGTP e TTP di grado UltraPure sciolti in acqua per biologia molecolare di elevata qualità. Un µL è sufficiente per una reazione standard di PCR da 50 µL.
Applicazioni
La miscela di dNTP è stata usata:
- nell'amplificazione mediante PCR di DNA genomico isolato da insetti,[1] funghi,[2] virus, uomo,[3]
- nella trascrizione inversa dell'RNA totale in cDNA.[4][4]
- come componente della miscela di amplificazione del DNA per la reazione a catena della polimerasi (PCR)[2][5][1]
- per le PCR di routine
- per il sequenziamento manuale e automatico del DNA
- per la sintesi del cDNA[6] nelle reazioni di marcatura
