Általános leírás

Monoclonal Anti-α-Tubulin (mouse IgG1 isotype) is derived from the B-5-1-2 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from an immunized mouse. Tubulin is the major building block of microtubules.
The TUBA4A (tubulin α 4a), is mapped to human chromosome 2q35. The gene codes for a member of the a -tubulin family, and contains 448 amino acids. TUBA4A belongs to the subfamily of tubulin, which is the major building block of microtubules. The α-subunit of tubulin has molecular weight of 50,000.

Egyediség

Monoclonal Anti-alpha-Tubulin antibody recognizes an epitope at the C-terminal end of the α-tubulin isoform in various organisms.
Recognizes an epitope located at the C-terminal end of the α-tubulin isoform in a variety of organisms.

Immunogén

Sarkosyl-resistant filaments from Strongylocentrotus purpuratus (sea urchin) sperm axonemes.

Alkalmazás

Monoclonal Anti-α-Tubulin antibody produced in mouse has been used:
  • In immunofluorescence Analysis
  • In western blotting/ Immunoblotting
  • For immunolabelling cells in electron microscopy

Biokémiai/fiziológiai hatások

Tubulin α 4a (TUBA4A) mutation is associated with amyotrophic lateral sclerosis (ALS).
α-Tubulin is a key regulator of cytoskeletal proteins. It mediates cellular developmental stages such as proliferation, migration, signalling and also maintains the shape of the cell. α-Tubulin controls trafficking, signaling and cellular tensegrity mediated by microtubules. The encoded protein is associated with the development and progression of cancer. α-Tubulin acetylation potentiates the metastatic property of breast cancer. Mutation in TUBA4A is associated with the development of various types of cancers, such as oral cancer, breast cancer, rectal cancer, lung cancer and prostate cancer. In addition, variation in the TUBA4A leads to sporadic amyotrophic lateral sclerosis (ALS).
Microtubules function as structural and mobile elements in mitosis, intracellular transport, flagellar movement and the cytoskeleton.
Tubulin α mediates the formation of cytoskeletal protein polymers. It is necessary for cell cycle and controls cellular movement and signaling. Tubulin α is known to maintain the shape of the cells. It is an active part of oncogenesis and progression of different types of cancer. Posttranslationally modified Tubulin α governs microtubule functions including cell trafficking and cellular tensegrity.

Tárolás és stabilitás

For continuous use, store at 2-8 °C for up to one month. For extended storage, the solution may be frozen in working aliquots. Repeated freezing and thawing, or storage in "frost-free" freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.

Cikkek

Microtubules of the eukaryotic cytoskeleton are composed of a heterodimer of α- and β-tubulin. In addition to α-and β-tubulin, several other tubulins have been identified, bringing the number of distinct tubulin classes to seven.
