Allgemeine Beschreibung

o-Phenylendiamin (o-PD) ist ein farbloses und nicht fluoreszierendes Molekül.[1]
o-Phenylendiamindihydrochlorid (OPD) ist ein chromogenes Substrat, das in Enzyme-linked-Immunosorbent-Assay(ELISA)-Verfahren eingesetzt werden kann, da es Meerrettichperoxidasekonjugate nutzt. Das Substrat produziert ein lösliches Endprodukt mit orange-brauner Farbe, das bei 450 nm spektrofotometrisch gemessen werden kann. Die OPD-Reaktion kann mit 3 N HCl oder 3 mol/l H2SO4 gestoppt und bei 492 nm gemessen werden.

Anwendung

o-Phenylendiamindihydrochlorid wird als Substrat für Meerrettichperoxidase-konjugierte sekundäre Antikörper im Enzymimmunoassay (ELISA) eingesetzt.[2][3][4]

Rekonstituierung

Lösen Sie eine Tablette in 0,05 M Phosphat-Citrat-Puffer, pH-Wert 5,0, um die gewünschte Konzentration zu erhalten (in der Regel wird eine OPD-Konzentration von 0,4 mg/ml verwendet). Geben Sie unmittelbar vor der Verwendung 40 μl frisches 30%iges Wasserstoffperoxid je 100 ml Substratpufferlösung hinzu. Die Tabletten sind einzeln in Folienpackungen verpackt.
