Beschreibung

General description

o-Phenylendiamindihydrochlorid (OPD) ist ein chromogenes Substrat, das in Enzyme-linked-Immunosorbent-Assay(ELISA)-Verfahren eingesetzt werden kann, da es Meerrettichperoxidasekonjugate nutzt. Das Substrat produziert ein lösliches Endprodukt mit orange-brauner Farbe, das bei 450 nm spektrofotometrisch gemessen werden kann. Die OPD-Reaktion kann mit 3 N HCl oder 3 mol/l H2SO4 gestoppt und bei 492 nm gemessen werden.

o-Phenylendiamindihydrochlorid ist ein Binukleophil, das durch die Kondensation mit α,β-ungesättigten Carbonylverbindungen und Aldehyden Benzimidazole bildet.

Application

o-Phenylendiamindihydrochlorid wurde als Substrat für die Meerrettichperoxidase verwendet, um Folgendes nachzuweisen:

• humanes Immunglobulin
• CTLA4Ig in Serumproben
• Albumin in Kulturmedien

Preparation Note

Lösen Sie eine Tablette 0,05 M Phosphat-Citrat-Puffer, pH 5,0, in der gewünschten Konzentration (in der Regel wird eine OPD-Konzentration von 0,4 mg/ml verwendet). Geben Sie unmittelbar vor der Verwendung 40 μl frisches 30%iges Wasserstoffperoxid pro 100 ml Substratpufferlösung hinzu. Die Tabletten sind einzeln in Folien verpackt.
